Well kind of wanted to avoid entering this since I have uploaded several such clips (9 with audio, 7 of which have subtitles, and 3 which are insert songs). I don't have a particularly strong interesting in protecting them, though out of those I do have a minor desire toward retention of insert songs (specifically those that are insert songs with no interruptions).

I believe on screenshots it's more of an issue of quality, as most are generated using over air broadcast episodes suffering compression artifacts, and the bulk of screencaps can be, well artistically worthless. A stitch depicting the entire character might be one thing, but a random frame out of an uninteresting shot is just going to be junk. For animation, I think that it's inevitable to have some compression artifacting with them and as long as it is moving the same degree of artifacting that would be horrible for a still is usually not an issue with animation.
As for rules, I feel that the ToS rule against uploading entire serialized manga would be the one that would be most applicable toward trying to upload an entire episode of anime. As the underlying reason against uploading the manga would be just as applicable to trying to upload anime. As for asking whether that applies to an entire skit of anime as opposed to the entire episode, I'll leave that for someone else to hammer out.
On the copyright/DMCA threat issue, it perhaps opens the door wider and thus easier for such an issue, but the site has always been open to such a threat simply for hosting artbooks and various flash videos that contain various copyrighted songs (see music_video tag).
Beyond an outright ban, reducing the lengths of the clips from anime might reduce the threat of having them on the site. While an agreed upon length might not be short enough to cover your example (would take a sub 18 second length), can certainly prevent lengths that would allow for OPs and EDs.
